Cr Williams joined the other mayors, including Brisbane Lord Mayor Graham Quirk on Friday.
Lord Mayor Quirk said the region had a lot to offer and now was the best time to start lobbying.
"We'll be advising the Australian Olympic Committee...the Queensland Government and Australian Government of our interest in having further discussions around this."
The mayors unanimously agreed to support the regional bid at the Council of Mayors board meeting.
Regional support for the bid moves SEQ one step closer to hosting the Games.
The mayors will go to Canberra in April to discuss federal support and prepare for discussions with the state government and Australian Olympic Committee.
A formal expression of interest for the 2028 Olympics is not required until January 2019, with a formal bid deadline later that year.
No funding has been allocated towards the bid, nor is it required at this stage.
Brisbane entered a bid for the 1992 Olympics but finished third behind Barcelona and Paris in the official ballot held in 1986.
The next Olympic Games will be in Rio de Janeiro in Brazil next year, with the 2020 Games to take place in Tokyo, Japan.
