The Northern Territory-based side came from behind in the second quarter to take the victory, courtesy of performances from Nathaniel Long, Garrick Ray, and Judah Philomac.
Young Guns player Nathan Ross kicked a game-high of four goals, while Jameson Billy, Sebastian Philomac, and Craig Philomac were in close support, bagging three goals each.
Half-forward Jayden Norman was outstanding for Buffaloes kicking two goals on debut, along with Tom Ketchup on the wing.
The Lake Nash side overcame a 13-point margin at quarter time to get back into the contest after its opponents made the most of an advantageous breeze early on.
But a poor restart from the third-place side let Young Guns straight back in the match, kicking three unanswered goals.
Ten goals in the second half was more than enough for the 2014 grand final runners-up to secure the all-important second spot on the ladder winning 15.8.98 to 6.5.41 over Buffaloes.
With both the Buffaloes and Rovers losing this round, the competition ladder is now set for the final series with first versus second for a grand-final spot, followed by third versus fourth in a knockout clash.
