A police spokesman said remains had been found at a home on Deighton Street, Mornington, and the caravan was destroyed.
A 39-year-old man and two children, aged four and seven, were believed to have been inside the caravan at the time, police said.
10am: Mount Isa Mayor Tony McGrady is returning from Brisbane because of the tragedy.
He was attending a planning conference in Brisbane hosted by Deputy Premier Jackie Trad.
Cr McGrady described the incident as "a terrible terrible time for Mount Isa"
"I think the community can do what it always does in times of tragedy...to just be there. We have to stand as one."
"A small community such as ours takes it harder, feels it harder, Cr McGrady said.
Deputy Premier Jackie Trad said the government would offer what assistance it could to the community.
"It is devastating, we are still obviously finding out the details in relation to this," she said.
"I know this is such a significant tragedy for [Cr McGrady]'s community.
"I want to reassure people the Queensland Government stands ready to help Mayor McGrady and the whole Mount Isa community in relation to this issue.
"Our hearts go out to those involved."
9.27am: A 39-Year-old man and two children aged four and seven are missing following a caravan explosion in Deighton Street.
Police are investigating the explosion since called to the address after 7am.
Police have set up an exclusion zone in Deighton Street between Enid, Stanley and Jones streets.
Police were called to the address just after 7am following reports of a possible gas explosion in a caravan, with a fire spreading to an adjoining house.
The remains of three people have been located at the scene.
Police have been advised that a 39-year-old man, and two children, aged four and seven, were believed to have been inside the caravan at the time.
An emergent situation was declared under the Public Safety Preservation Act at 7.30am with an exclusion zone in Deighton Street between Enid, Stanley and Jones streets.
Scientific and Scenes of Crimes officers are at the scene and police investigations are continuing.
Nearby residents self-evacuated and there is no further information available at this time.
8.50am: THREE people are missing after an explosion in Deighton Street this morning.
The explosion could be felt across Mount Isa when it happened at about 7am, Tuesday.
Mount Isa Police Inspector Trevor Kidd described the explosion as a "serious situation" that happened near Ann Street.
Deighton Street had been cordoned under the Public Safety Preservation Act, Inspector Kidd said.
"We have three people unaccounted for.
"All emergency services are on the scene.
"It's a serious situation and we are asking people to keep clear," he said.
