Runners will hit the road at 6pm on June 16 at Duchess, and have until 8am the following morning to arrive in Mount Isa.
Almost 100 people took park in the 96-kilometre marathon last year.
Event organiser Alison Whitehead said the run is a fun way to give back to the community.
“You can enter a team of up to 10 people,” Ms Whitehead said.
“We get a few people that want to do the whole 100 by themselves, but most people enter in a team.
“It is something out of the ordinary to do with your friends, it is a run in the dark down a bush road – something you wouldn’t normally do” she said.
“The police do a really great job of keeping everyone safe and they usually enter a team as well.”
Ms Whitehead said funds raised this year would go towards mental health in Mount Isa.
“Something needs to be done because a lot of people in the community are struggling,” Ms Whitehead said.
“I think we will give the money to Mount Isa Mental Health Network and they can use it for mental health week in October.
“Last year we raised about $5000,” she said.
There are plenty of games and checkpoints along the way to keep runners and spectators awake.
